ISSX President’s Message By John O. Miners, ISSX President I would like to take this opportunity to urge you to attend the 20th North American ISSX Meeting, which will be held in Orlando in just a couple of weeks. Hartmut Derendorf and Stephan Schmidt, the Meeting Cochairs, and their colleagues on the Meeting Organizing Committee have put together an excellent scientific program. The meeting begins on Sunday, October 18 with four short courses, followed by the Opening Keynote Lecture and the Welcome Reception. The plenary lectures and ten symposia scheduled between Monday, October 19 and Thursday, October 22 address a diverse range of topics that include sources of variability in PK and PD, transporters, drug-drug interactions, drug design and development, drug and chemical toxicity (including environmental exposure to drugs and personal care products), and biologics. These symposia are complemented with poster sessions, industry-sponsored symposia, a comprehensive trade exhibition, and a lecture from the
recipient of the North American Scientific Achievement Award (in honor of Ronald W. Estabrook), Leslie Benet. As noted in a previous newsletter, ISSX Council is keen to increase the profile of new investigators in ISSX and enhance their input into the activities of the Society. In this regard, a dedicated session of oral presentations of selected abstracts from new investigators and a New Investigators Forum about career and leadership development are included in the meeting program. New investigators themselves have been instrumental in the planning and organization of both sessions. The 20th North American ISSX Meeting promises to be rewarding both scientifically and socially. The venue offers the opportunity to enjoy the local theme parks and other attractions with your family and/or your colleagues. A free afternoon has been scheduled on Tuesday, October 20 and especially reduced-price tickets to several local attractions
are available to meeting attendees. My predecessor Bill Smith and I have emphasized in previous newsletters that increasing internationalization and scientific outreach are core objectives of ISSX. The Society has established close ties with the Japanese Society for the Study of Xenobiotics (JSSX) having run joint meetings with JSSX in 2005 and 2014. (A third joint meeting with JSSX is scheduled to be held in Hawaii in 2020.) There was also a joint meeting with Microsomes and Drug Oxidations in 2012. ISSX collaborates closely with the Chinese Society for the Study of Xenobiotics (CSSX), and CSSX has been instrumental in the planning and organization of several Asia Pacific ISSX meetings held in China. “The affinity optimization of drug candidates is a major goal in drug." (p. 3). So starts the first chapter and sets the scene for the remainder of the book. It describes the theoretical background and unites this with traditional, experimental methodology and modern, computational approaches. The application to, and understandings learnt from, actual drug development programs are included as case studies with details of potential solutions that may be applicable to other ongoing drug optimization problems. The book is divided into three sections: the first two, not unexpectedly, are termed, “Thermodynamics” and “Kinetics,” with the last segment titled “Perspective,” summarizing the situation and evaluating the practicality of this knowledge. The seven chapters in the first section are entitled: “The binding thermodynamics of drug candidates,” “van’t Hoff based thermodynamics,” “Computation of drug-binding thermodynamics,” “Thermodynamics-guided optimizations in medicinal chemistry,” “From molecular understanding to structure – thermodynamic relationships, the case of acetylcholine binding proteins,” “Thermodynamics in lead optimization,” and “Thermodynamic profiling of carbonic anhydrase inhibitors.” This is followed by another eight chapters: “Drug-target residence time,” “Experimental methods to determine binding kinetics,” “Challenges in the medicinal chemical optimization of binding kinetics,” “Computational approaches for studying drug binding kinetics,” “The use of structural information to understand binding kinetics,” “Importance of drug-target residence time at G protein-coupled receptors – a case for the adenosine receptors,” “Case study: Angiotensin receptor blockers (ARBs),” and “The kinetics and
thermodynamics of Staphylococcus aureus Fab1 inhibition.” The final conclusory part contains only one chapter: “Thermodynamics and binding kinetics in drug discovery.” Thirty-four authors have participated in this project, which is enumerated as Volume 65 in the, “Methods and Principles in Medicinal Chemistry” series that has been running since 1993. The sixteen chapters have copious sub-headings that break the text into readable portions, contain graphs (many in color), schemes, diagrams, and both mathematical and chemical equations. The end of the book features a list of pertinent references that enable access to more detailed information if required. The book has a short, four-page index with only about 200 entries, but this is sufficient to navigate the volume. This offering is an ideal reference tome for those who are interested in, or work within, this important field of drug development and design.

Leslie Benet to Receive Scientific Achievement Award The International Society for the Study of Xenobiotics proudly announces that Professor Leslie Z. Benet is the winner of the 2015 North American Scientific Achievement Award in Honor of Ronald W. Estabrook. This award is sponsored by XenoTech, LLC. Leslie Z. Benet is a Professor of Bioengineering and Therapeutic Sciences at the University of California, San Francisco (UCSF) in San Francisco, California, United States. He has been a member of the UCSF School of Pharmacy faculty since 1969 and chaired the school's Department of Biopharmaceutical Sciences from 1978 to 1998. He received an A.B. (English), B.S.Pharm., and M.S.Pharm. from the University of Michigan and his Ph.D. from the University of California. Dr. Benet is listed by Web of Science as one of the most highly-cited pharmacologists worldwide. His most recent work has addressed the interplay of metabolic enzymes and transport proteins as related to oral bioavailability and hepatic elimination, with particular emphasis on immunosuppressive, anti-cancer, anti-AIDS, and antiinfective drugs. He co-developed the Biopharmaceutics Drug Disposition Classification System, which has become a widely-used clinical tool for determining drug bioavailability and disposition.
The North American Scientific Achievement Award will be presented at the 20th North American ISSX Meeting at the awards presentation session on Monday, October 19. ISSX Awards Committee Chairman, Dr. Geoffrey T. Tucker, Emeritus Professor of Clinical Pharmacology at the University of Sheffield, United Kingdom, will present the honor to Dr. Benet with ISSX President Dr. John O. Miners. Dr. Benet will then deliver his award lecture. Please join ISSX in congratulating Dr. Benet on this outstanding achievement.

One of Dr. Benet’s supporting nominators for the award stated, “Just as Dr. Estabrook significantly furthered the understanding of how the body metabolized drugs during his time, Les has uncovered many new principles related to drug metabolism and transport, and importantly to pharmacokinetics. He has had a marked impact on the clinical importance of drug metabolism, transport, and pharmacokinetics.”

Dr. Benet was a founder and editor of the Journal of Pharmacokinetics and Biopharmaceutics and an associate editor of Pharmacology and Therapeutics. He is a Fellow of the Academy of Pharmaceutical Sciences and Research, the American Association of Pharmaceutical Scientists (AAPS), and the American Association for the Advancement of Science (AAAS). He was President of the Academy of Pharmaceutical Sciences in 1985 and in 1986 he was the co-founder and first President of AAPS. His service to ISSX includes leadership positions as a member of Council (1991–1995) and service as the Society’s Treasurer (1998–2000). Dr. Benet has been a member of ISSX for more than 29 years.

for Xenobiotic Research (ISXR), which has been registered as a legal entity in India. Murali Subramanian is the inaugural President of ISXR. Murali will be supported by a Board of Directors that includes Ravikanath Bhamidipati as Secretary and Abhay Sagamwar as Treasurer. It is anticipated that ISXR will become an affiliate organization of ISSX, and planning is underway for an ISSX-sponsored meeting of ISXR in India in the second half of 2016.

ISSX Council and the ISSX Office strive to ensure that the Society provides high-quality services to its members, particularly scientific meetings, and scientific leadership in the area of xenobiotic metabolism and disposition more generally. In this regard, it is important that ISSX adapts to remain relevant and influential in the current challenging environment that faces scientists in academia and industry. During the course of the 20th North American ISSX Meeting, Council will develop a strategic plan that will allow us to fulfil our core objectives over the next three years. DMPK Society Forms in India Internationalization and organizing scientific educational activities within India has been discussed by ISSX Council since 2008. In early 2013, under the leadership of ISSX President Bill J. Smith, Council advanced the development of a permanent association of scientists in India interested in promoting the understanding of the interactions of medicines and chemicals with living systems. Following his attendance at a meeting in Hyderabad, India in March 2013, Dr. Smith discussed this concept with Dr. Sandhya Mandlekar, Head of Pharmaceutical Candidate Optimization at Biocon Bristol-Myers Squibb Research Center in Bangalore, India. He shared his observation that India bore many similarities to the DMPK landscape in China when ISSX began increasing activities there and one of the things that helped ISSX significantly in China was the existence of the Chinese Society for the Study of Xenobiotics. Dr. Smith asked Dr. Mandlekar whether it might be possible to develop a group in India to use as a platform to increase promotion of the science there. There was significant interest amongst DMPK scientists in India. ISSX was fortunate to have Dr. Murali Subrmanian, Lead Investigator at Biocon Bristol-Myers Squibb Research Centre in Bangalore, attend the 2013 Council meeting in Toronto. During this meeting he outlined the landscape and provided suggestions on where ISSX could contribute to continuing education in India. Plans were made for the participation of ISSX in several sessions at the 2014 NIPER DMPK Symposium meeting. NIPER is the National Institute of Pharmaceutical Education and Research, an autonomous body set up under the aegis of the Department of Pharmaceuticals, Ministry of Chemicals and Fertilizers, Government of India.

ISXR will work to organize a scientific conference in 2016 and recruit colleagues within India as meeting organizing committee members. Future activities will include a membership drive consisting of targeted emails and social media. The short term goal is to host two symposia in the major cities of Mumbai and Bangalore along with a half-day vendor-sponsored modeling workshop in 2016. The long term goal is a three-day meeting each year beginning in 2017. ISSX congratulates everyone involved in the organization of ISXR as a national scientific society in India. We will be certain to maintain awareness of their activities amongst the ISSX membership and wish them much success in their endeavors.
