Trước đây, một vài loại trái cây đã được chứng minh là có hoạt tính chống oxy hóa mạnh. Trong bài báo này, chúng tôi báo cáo kết quả đánh giá về hoạt tính chống oxy hóa của 22 loại rau phổ biến , trà xanh, trà đen được xác định theo phương pháp ORAC assay (khả năng hấp thụ gốc oxy hóa), sử dụng 3 tác nhân oxy hóa khác nhau, bao gồm: tác nhân sinh gốc tự do peroxyl, tác nhân sinh gốc tự do hydroxyl và ion Cu2+ một kim loại chuyển tiếp. Căn cứ trên khối lượng tươi, tỏi có hoạt tính chống oxy hóa cao nhất (tính theo đơn vị tương đương µmol of Trolox /g) đối với gốc tự do peroxyl (19.4), tiếp theo là cải xoăn (17.7), rau spi-nách (12.6), cải Brussel, rau mầm cỏ linh lăng, súp lơ xanh, củ cải đường, ớt chuông, hành, ngô, cà tím (9.8- 3.9), xúp lơ răng, khoai tây, khoai tây ngọt, cải bắp, rau diếp, bí vàng, xà lách, cần tây và dưa chuột (3.8-0.5); cải xoăn có hoạt tính chống oxy hóa tốt nhất với các gốc tự do hydroxyl, tiếp theo đó là cải Brussel, rau mầm cỏ linh lăng, củ cải ngọt, rau spi-nách, súp lơ xanh và các loại rau củ khác. Trà xanh và trà đen có hoạt tính chống oxy hóa vượt trội so với tất cả các loại rau củ trên. Tuy nhiên, trà cũng cho thấy tác dụng gây tress oxy hóa (pro- oxidant) khi có mặt tác nhân Cu2+ , điều này không thấy ở bất kỳ nghiên cứu nào trên rau củ.

Tác dụng của dự phòng của hoa quả và rau xanh đối với các loại bệnh ung thư, bệnh tim mạch, bệnh mạch máu não, được cho là do vai trò của rất nhiều chất oxy hóa, đặc biệt là các vitamin có hoạt tính chống oxy hóa như vitamin C, α- tocopherol chứa trong các loại rau quả này (Ames, 1983; Gey, 1990; Geyet al., 1991; Riemersma et al., 1989; Stahelin et al.,1991a,b; Steinberg et al., 1989, 1991; Willett, 1994b). Tuy nhiên, phần lớn tác dụng chống oxy hóa của các loại rau quả này có thể đến từ các hợp chất khác thay vì vitamin C, vitamin E hay β-caroten. Ví dụ một số flavonoid được tìm thấy ở trong bữa ăn có hoạt tính chống oxy hóa (Bors and Saran, 1987; Bors etal., 1990; Hanasaki et al., 1994). Phòng thí nghiệm của chúng tôi đã báo cáo về một số một số loại rau củ thường gặp có hoạt tính chống oxy hóa cao nhưng không thể giải thích dựa trên hàm lượng vitamin C của chúng (Wang et al., 1996). Chúng tôi cũng tìm thấy một số flavonoid có hoạt tính chống oxy hóa đối với các gốc peroxyl mạnh hơn nhiều so với vitamin E, vitamin C và gluthation (Cao et al., in press). Hoạt tính chống ung thư của rau xanh đã được phát hiện trên các nghiên cứu trên tế bào (Maeda et al., 1992) và trên động vật (Belman, 1983; Bingham, 1990; Bresnick et al.,1990; Maltzman et al., 1989; Stoewsand et al., 1988; Stoewsand et al., 1989; Wattenberg and Coccia, 1991). Có mối quan hệ tỷ lệ nghịch có ý nghĩa giữa việc sử dụng hoa quả và rau xanh với tỷ lệ tử vong do bệnh tim mạch và bệnh mạch máu não (Acheson and Williams, 1983; Armstrong et al., 1975; Burr and Sweetnam, 1982; Phillips et al., 1978; Verlangieri et al.,1985). Sử dụng lượng lớn hoa quả và rau ỏ cả người ăn chay và không ăn chay đều làm giảm huyết áp (Ascherio et al., 1992; Sacks and Kass, 1988).

Hóa chất: β-Phycoerythrin (β-PE) có trong tảo Porphydiumcruentum được mua của công ty Sigma (St. Louis, MO). β-PE được sử dụng trong những thí nghiệm này thường mất 90% huỳnh quang trong vòng 30 phút khi có sự hiện diện của 4 mmol/L AAPH. AAPH được mua từ công ty Wako Chemicals USA Inc. (Richmond, VA). 6-Hydroxy 2,5,7,8tetramethyl- chroman-2-carboxylic acid (Trolox) được mua từ công ty Aldrich (Milwaukee, WI).

Trà và rau: 22 loại rau được mua thành 3 đợt riêng rẽ ở các siêu thị địa phương. Các loại rau bao gồm: tỏi, cải xoăn, rau spi-nách, cải Brussel, rau mần cỏ linh lăng, súp lơ xanh, củ cải đường, chuông ớt, hành, ngô, cà tím, súp lơ trắng, khoai tây, khoai tây ngọt, búp cải, rau diếp, đậu tây, cà rốt, bí vàng, xà lách, cần tây, dưa chuột. TRà xanh được sử dụng trong các nghiên cứu này là trà Chin Chu oriental blend (hỗn hợp phương Đông). Trà đen (tất cả trà đen là trà lên men) là ở dạng bột khô được cung cấp bởi hiệp hội Tea Trade Health Research.

Trà đen được hòa tan hoàn toàn trong nước khử ion với tỷ lệ 5mg/ml và sử dụng phương pháp ORAC assay để định lượng trực tiếp sau khi pha loãng bằng đệm phosphat 75mM, pH 7.0. Trà xanh được ủ trong nước khử ion trong vòng 30 phút ở 95-100 oC với tỷ lệ 1:60 khối lượng/thể tích. Phần sử dụng để chế biến làm thức ăn của các loại rau được cân , đồng nhất bằng cách xay nhuyễn sau khi thêm vào 1 thể tích nước khử ion với tỷ lệ 1:2 khối lượng/thể tích. Trà xanh sau khi ủ, rau xanh nhuyễn đồng nhất được đem đi ly tâm ở tốc độ 34000/phút trong 30 phút ở 4 oC. Phần bã (phần tan trong nước) được tách và sử dụng trực tiếp trong phương pháp ORAC assay sau khi pha loãng thích hợp với đệm phosphat. Phần cơ chất sa lắng sau khi ly tâm (phần không tan trong nước) được rửa 2 lần với nước khử ion và sau đó được chiết với aceton (1:4 w/v) bằng cách lắc ở nhiệt độ phòng trong vòng 30 phút. Aceton đã được sử dụng trong các lab của chúng tôi (Wang et al., 1996) và những người khác (Daniel et al., 1989; Mass et al., 1991) để chiết tách các chất oxi hóa từ bã trái cây. Dịch chiết

Xác định khả năng hấp thụ gốc tự do bằng phương pháp tự động: được tiến hành trên máy COBAS FARA II phân tích lý tâm quang phổ huỳnh quang (hãng Roche Diagnostic System Inc., Branchburg, NJ) with fluorescent filters (ex, 540 nm; em, 565 nm). Phương pháp tiến hành dựa trên mô tả trong nghiên cứu của Cao và đồng nghiệp (Cao et al., 1993) cũng như những thay đổi khi thao tác trên máy COBAS FARA II (Cao et al., 1995). Tóm lại trong hỗn hợp thử cuối cùng, β- PE (16.7 nM) được sử làm đích tấn công của các gốc tự do (hoặc chất oxi hóa) với (AAPH) một chất sinh gốc peroxyl (ROO.) (ORACROO assay),, (ii) Cu2+-H2O2 [H2O2, 0.3%; Cu2+ (CuSO4), 9 µM] là nguồn sinh gốc hydroxyl (OH.) (ORACOH assay). (iii) Cu2+ (dạng CuSO4) (18 µM) như là một chất oxy hóa kim loại chuyển tiếp (ORACCu assay). Trolox được sử dụng như một chất chứng chuẩn. Dung dịch gốc ở nồng độ 0.1 mM sẽ ổn định tối thiểu 1 tháng ở -80 o C. Máy phân tích được lập trình để ghi lại huỳnh quang của β-PE mỗi 2 phút 1 lần sau khi AAPH, Cu2+-H2O2 hoặc Cu2+ được thêm vào. Kết quả cuối cùng được tính toán dựa trên sự chênh lệch diện tích dưới đường cong biểu diễn sử phân hủy β-PE ở mẫu trắng và mẫu thử, kết quả biểu diễn µmol Trolox/ gram trà hoặc rau, ngoại trừ trường hợp khi Cu2+ sử dụng đơn độc (ví dụ không có H2O2) làm tác nhân oxy hóa. Khi có mặt Cu2+ đơn độc, Trolox không thể sử dụng như một tác nhân chống oxy hóa vì lúc này Trolox thể hiện tính pro-oxidant (are chemicals that induce oxidative stress- wiki) có thể gây stress oxi hóa do có mặt Cu2+ (Cao and Cutler, 1993). Vì vậy kết quả của ORACCu assay trong trường hợp này sẽ được tính toán theo công thức (diện tíchthử - diện tíchtrắng)/ diện tíchtrắng và biểu diễn dưới dạng đơn vị của chất oxy hóa trong đo 1 đơn vị tương đương với hoạt tính oxy hóa làm tăng 100 % diện tích dưới đường cong phân hủy β-PE. Giá trị âm của ORACCu chỉ dấu có phản ứng pro-oxidant gây ra bởi Cu2+.

Khả năng hấp thụ gốc oxy hóa của các loại rau và trà xanh được tính bằng tổng khả năng hấp thụ gốc oxy hóa của dịch chiết nước và dịch chiết aceton. Trọng lượng khô của rau được xác định sau khi sấy ở 40 oC trong 1 tuần.

KẾT QUẢ
Hoạt tính chống lại các gốc oxy hóa peroxyl (ORACROO.) của 22 loại rau củ phổ biến, trà xanh, trà đen thể hiện ở bảng 1. Căn cứ trên khối lượng tươi, trong các loại rau của thì tỏi và cải xoăn đứng đầu về khả năng hấp thụ các chất oxy hóa ROO. trong 22 loại rau. Rau spi-nách, cải Brussel, rau mần cỏ linh lăng, súp lơ xanh, củ cải đường, ớt chuông, hành, ngô và cà tím có giá trị ORACROO. rơi vào nhóm trung bình (3.9 -12.6). Súp lơ trắng, khoai tây, khoai tây ngọt, bắp cải, rau diếp, đậu tây, cà rốt, bí vàng, xà lách, cần tây và dưa chuột là nhóm có chỉ số ORACROO. thấp nhất trong các loại rau được đánh giá. Dựa trên khối lượng tươi, cải xoăn có khả năng hấp thụ các gốc oxy hóa hydroxyl tốt nhất, tiếp theo đó là cải Brussel, rau mần cỏ linh lăng, củ cải ngọt, rau spi-nách, súp lơ xanh, ngô, đậu tây, bắp cải, rau diếp, cà tím, súp lơ xanh, bí vàng, tỏi, khoai tây, khoai tây ngọt, cà rốt, xà lách, ớt chuông, hành, cần tây và dưa chuột. Trà xanh cho thấy hoạt tính pro-oxidant ( hoạt tính ORACCu âm ) khi có mặt Cu2+ đơn độc ( không có mặt H2O2) (Bảng 1). Đây là hoạt tính prooxidant gây ra bởi ion Cu2+, tuy nhiên điều này không quan sát thấy ở bất kỳ loại rau củ nào được khảo sát trong nghiên cứu. Căn cứ trên khối lượng tươi, tỏi có hoạt tính chống oxy hóa cao nhất đối với tác nhân Cu2+ ( hoạt tính ORACCu). Tuy nhiên rau spi-nách sẽ có hoạt tính chống oxy hóa cao nhất đối với tác nhân Cu2+ nếu hoạt tính này căn cứ trên khối lượng khô, tiếp theo đó là xà lách, súp lơ xanh và các loại rau khác.

Mức điểm chống oxy hóa của từng loại rau thể hiện ở Bảng 1 được tính toán đơn giản bằng việc cộng 3 gá trị: ORACROO. (đơn vị tương đương 1 µmol Trolox), ORACOH. (đơn vị tương đương 1 µmol Trolox), ORACCu (103 unit), căn cứ trên khối lượng tươi của rau củ. Một (tương đương) nano mol của Trolox được tính toán trong phép phân tích ORACROO. Và 1 đơn vị

ORACCu được tính toán trong phép định lượng ORACCu đều là biểu diễn cùng 1 giá trị diện tích chênh lệch dưới đường cong phân hủy β-PE giữa mẫu trắng và mẫu thử, cái mà được sử dụng để tính toán trong phép đo định lượng ORAC. Vì giá trị điểm hoạt tính chống oxy hóa của các loại rau đối với gốc peroxyl (ORACROO.) lớn hơn nhiều so với giá trị này đối với các gốc hydroxyl (ORACOH.) và tác nhân Cu2+ (ORACCu) nên, đối với 2 tác nhân sau, các giá trị ORAC của các loại rau không được phân hạng như đối với giá trị ORACROO. . Giá trị điểm chống oxy hóa không áp dụng đối với trà do ban đầu chúng đã ở dạng khô, không phải dạng tươi.

Hoạt tính chống tác nhân oxy hóa của trà và các loại rau củ được xác định ở nhiều nồng độ dịch chiết, nguyên nhân là do tác động pro-oxidant gây ra bởi Cu đối với một số chất chống ở oxy hóa chỉ quan sát thấy ở mức nồng độ tương đối cao (Cao and Cutler, 1993). Kết quả thể hiện ở biểu đồ 1 cho thấy khi có mặt Cu2+ đơn độc ( không có mặt H2O2), trà hoạt động như 1 chất pro-oxidant ở tất cả các nồng độ và hoạt tính pro-oxidant tăng khi tăng nồng độ của trà. Tuy nhiên trong các thí nghiệm với các loại rau bao gồm rau spi-nách, tỏi, súp lơ xanh, tất cả đều hoạt động như chất chống oxy hóa đối với tác nhân Cu2+ và hoạt tính của chúng tăng tỷ lệ thuận với nồng độ.

Phương pháp đo khả năng hấp thụ các gốc oxy hóa (ORAC assay) được phát triển gần đây bởi Cao và các cộng sự (Cao et al., 1993, 1995) cung cấp một phương pháp riêng biệt và mới để đánh giá hoạt tính chống oxy hóa của các hợp chất đa dạng và các mẫu sinh học. Phương pháp này ưu điểm vượt trội so với các phương pháp tương tự khác vì 2 lý do. Đầu tiên, hệ thống đánh giá hoạt tính ORAC sử dụng kỹ thuật diện tích dưới đường cong vì vậy đã kết hợp trong 1 phép định lượng có thể phản ánh được cả thời gian và mức độ ức chế các gốc tự do của các tác nhân chống oxy hóa (Cao etal., 1995). Các phương pháp tương tự khác (Ghiselli et al., 1994; Glazer, 1990; Miller et al., 1993; Wayner et al., 1985; Whitehead et al., 1992) đánh giá thời gian ức chế ở một mức độ ức chế nhất định hoặc xác định mức độ ức chế trong một khoảng thời gian nhất định làm nền tảng cho việc định lượng kết quả. Thứ hai, nhiều chất tạo sinh gốc tự do và chất oxy hóa khác nhau có thể sử dụng trong phương pháp định lượng ORAC này. Điều này là rất quan trọng vì việc đo hoạt tính chống oxy hóa của các mẫu sinh học phụ thuộc vào gốc tự do hay chất oxy hóa nào được sử dụng trong phép đo (Cao et al., 1996a,b).

Căn cứ trên cách tính toán trên, cải xoăn, củ cải đường, ớt chuông, cải Brussel, súp lơ xanh, rau spi-nách, khoai tây, ngô có sẽ cung cấp 1 lượng dang kể các chất có hoạt tính chống các gốc oxy hóa peroxyl trong bữa ăn nếu các loại rau củ này được sử dụng thường xuyên. Tần suất sử dụng mỗi loại rau trên có thể là một tác nhân khác để xác định loại rau củ nào trong chế độ ăn thôn thường sẽ đóng góp nhiều nhất vào việc khử các gốc oxy hóa (hoạt tính ORAC).
Gốc peroxyl (ROO. ) là gốc tự do phổ biến được tìm thấy trong cơ thể người và được sử dụng trong các phép đo hoạt tính chống oxy hóa (Wayner et al., 1985; Glazer, 1990; Cao et al., 1993,1995; Ghiselli et al., 1994). Nó kém hoạt động hơn gốc tự do hydroxyl (OH.) và vì vậy có thời gian bán thải dài hơn với đơn vị tính là giây thay vì nano giây (Grisham, 1992). Trong nghiên cứu này, Cu2+-H2O2 ( tác nhân sinh OH.) và Cu2+ đơn độc đã được sử dụng đểđánh giá hoạt tính chống oxy hóa của 1 loại trà xanh và 22 loại rau củ. Đa số gốc tự do OH. được cho là được tạo ra trong in vivo từ các kim loại khử - phụ thuộc H2O2 , ngoại trừ bất thường do tiếp xúc với phóng xạ ion hóa. Trong in vitro, các kim loại có thể sử dụng là titan, đồng, sắt , cô ban nhưng lựa chọn tốt nhất để kích thích sự hình thành các gốc tự do OH. trong in vivo có vẻ là sắt và kém hơn 1 chút là Cu2+H2O2 và Cu2+ đơn độc – thường xuyên để tạo ra các tổn thương oxy hóa lên protein và acid nucleotid (Parthasarathy et al., 1989; Sato et al., 1992). Phép đo hoạt tính ORACCu sử dụng Cu2+-H2O2 làm tác nhân sinh OH. sử dụng để đánh giá các hợp chất như manitol, glucose, acid uric (ở nồng độ sinh lý), protein, phức chelat của kim loại chuyển tiếp nhưng không phải là các hợp chất như acid ascorbic vì các chất này sẽ phản ứng trực tiếp với đồng để tạo ra các chất phản ứng.

Phép đo hoạt tính ORACCu sử dụng Cu đơn độc không chỉ đo lường hoạt tính chống oxy hóa ( giá trị ORACCu dương) mà còn xác định hoạt tính pro-oxidant gây ra bởi kim loại chuyển tiếp ( giá trị ORACCu âm) của các hoạt chất như acid ascorbic (Cao et al., 1993),và flavonoid (Cao et al., in press). Ở điểm này, chúng tôi không có những chỉ dấu về việc chất sinh gốc tự do nào sẽ là tốt nhất cho việc ước tính khả năng chống oxy hóa của các loại rau củ. Tuy có thể việc phân hạng “ điểm chống oxy hóa” , cái mà được áp dụng vào phân hạng hoạt tính chống oxy hóa của từng loại rau đối với 3 tác nhân khác phản ứng khác nhau, có thể sẽ cung cấp cho chúng tôi thông tin về vấn đề trên. Chúng tôi tính toán “ điểm chống oxy hóa” của từng loại rau trong nghiên cứu bằng việc cộng 3 giá trị: ORACROO. (đơn vị tương đương 1 µmol Trolox), ORACOH. (đơn vị tương đương 1 µmol Trolox), ORACCu (103 unit), căn cứ trên khối lượng tươi của rau củ. Một (tương đương) nano mol của Trolox được tính toán trong phép phân tích

Kết quả của chúng tôi chứng minh rõ ràng rằng tất cả các loại rau củ được đánh giá trong nghiên cứu đều có tác dụng chống oxy hóa không chỉ với gốc peroxyl mà với cả gốc hydroxyl và kim loại chuyển tiếp Cu2+ , tuy nhiên khả năng chống oxy hóa đối với từng tác nhân thay đổi đáng kể giữa các chủng loại rau củ khác nhau. Một kết quả quan trọng được phát hiện đó là những loại rau củ trong nghiên cứu ( kể cả các loại quả như dâu tây, dữ liệu không công bố) đều hoạt động như một chất chống oxy hóa khi tác nhân kim loại chuyển tiếp được sử dụng trong phép định lượng ORAC và tác dụng này tăng lên tỷ lệ thuận với nồng độ. Các kim loại chuyển tiếp xúc tác cho hoạt tính pro-oxidant của acid ascorbic (Beach và Giroux, 1992)) và α-tocopherol (Iwatsuki et al., 1995; Maiorino et al., 1993; Yoshida et al., 1994) đã được miêu tả. Sử dụng Cu2+-H2O2 trong phép định lượng ORAC, cho thấy acid ascorbic và Trolox – một dẫn chất tan trong nước của α-tocopherol (ở nồng độ tương đối cao) hoạt động như các chất pro-oxidant (Cao and Cutler, 1993). ORACROO. Và 1 đơn vị ORACCu được tính toán trong phép định lượng ORACCu đều là biểu diễn cùng 1 giá trị diện tích chênh lệch dưới đường cong phân hủy β-PE giữa mẫu trắng và mẫu thử, cái mà được sử dụng để tính toán trong phép đo định lượng ORAC. Vì giá trị điểm hoạt tính chống oxy hóa của các loại rau đối với gốc peroxyl (ORACROO.) lớn hơn nhiều so với giá trị này đối với các gốc hydroxyl (ORACOH.) và tác nhân Cu2+ (ORACCu) nên, đối với 2 tác nhân sau, các giá trị ORAC của các loại rau không được phân hạng như đối với giá trị ORACROO. .

Trà xanh và trà đen có hoạt tính chống gốc oxy hóa peroxyl (ORACROO.) cao hơn rất nhiều so với tất cả các loại rau và trái cây chúng tôi đã từng nghiên cứu. Giá trị ORACROO. căn cứ trên khối lượng khô gấp từ 4.5 -6 lần so với cả xoăn và dâu tây (Wang et al., 1996). Giá trị ORACOH. Tính trên khối lượng khô của trà xanh chỉ bằng 58% so với cải xoăn. Hoạt tính hấp thụ gốc tự do hydroxyl của trà xanh bị giảm khi có mặt Cu2+ trong khi giá trị ORACCu của trà xanh là giá trị âm chỉ ra trà xanh thể hiện tác dụng pro-oxidant khi có mặt Cu2+. Rõ ràng dường như có một số hoạt chất trong trà xanh có thể hấp thụ các gốc hydroxyl và một số gốc phản ứng khác được tạo ra từ phản ứng giữa Cu2+ và H2O2. Một số hợp chất trong trà rõ ràng có thể tạo ra các gốc phản ứng trực tiếp từ phản ứng giữa các thành phần trong trà, Cu2+ và O2 khi Cu2+-H2O2 được sử dụng làm chất sinh các gốc hoạt động trong phép đo ORAC. Và cũng có thể một số hợp chất trong trà như flavonoid (Cao et al., in press) có thể đóng đồng thời 2 vai trò đối lập tại cùng một thời điểm. Tuy nhiên, hoạt tính pro-oxidant gây ra bởi các kim loại chuyển tiếp đối với trà, acid ascorbic, α-tocopherol có thể không đóng vai trò quan trọng trong in vivo nơi mà các kim loại chuyển tiếp phần lớn hiếm gặp, ngoại trừ trong những bệnh cảnh có liên quan đến quá tải nồng độ ion kim loại. Tóm lược lại, hoạt tính chống oxy hóa của 22 loại rau củ, trà xanh, trà đen đã được xác định bằng phương pháp đo khả năng hấp thụ các gốc oxy hóa, sử dụng 3 tác nhân oxy hóa khác nhau, bao gồm: tác nhân sinh gốc tự do peroxyl, tác nhân sinh gốc tự do hydroxyl và ion Cu2+ một kim loại chuyển tiếp. Cải xoăn có hoạt tính hấp thụ các gốc tự do peroxyl tốt nhất căn cứ trên khối lượng khô. Trà xanh và trà đen có giá trị ORACROO. cao hơn rất nhiều so với bất kỳ loại rau nào trong nghiên cứu. Tuy nhiên trà thể hiện hoạt tính pro-oxidant gây ra bởi ion Cu2+, điều này cũng đã được quan sát thấy ở các chất chống oxy hóa khác như acid ascorbic, α-tocopherol, điều này không quát sát thấy trong các phân tích trên các loại rau củ. Vì vậy việc bổ sung các chất chống oxy hóa tự nhiên thông qua chế độ ăn cân bằng giữa rau củ và trái cây là cách hiệu quả trong việc giúp cơ thể chống lại những stress oxy hóa.

Previously, some fruits were shown to contain high antioxidant activities. In this paper, we report the antioxidant activities of 22 common vegetables, one green tea, and one black tea measured using the automated oxygen radical absorbance capacity assay with three different reactive species: a peroxyl radical generator, a hydroxyl radical generator, and Cu2+, a transition metal. Based on the fresh weight of the vegetable, garlic had the highest antioxidant activity (µmol of Trolox equiv/g) against peroxyl radicals (19.4) followed by kale (17.7), spinach (12.6), Brussels sprouts, alfalfa sprouts, broccoli flowers, beets, red bell pepper, onion, corn, eggplant (9.8-3.9), cauliflower, potato, sweet potato, cabbage, leaf lettuce, string bean, carrot, yellow squash, iceberg lettuce, celery, and cucumber (3.8-0.5); kale had the highest antioxidant activity against hydroxyl radicals followed by Brussels sprouts, alfalfa sprouts, beets, spinach, broccoli flowers, and the others. The green and black teas had much higher antioxidant activities against peroxyl radicals than all these vegetables. However, the tea also showed a prooxidant activity in the presence of Cu2+, which was not found with any of the vegetables studied. Keywords: Antioxidant; free radical; tea; vegetable INTRODUCTION
Consumption of fruits and vegetables has been associated with lower incidence and lower mortality rates of cancer in several human cohort and case-control studies for all common cancer sites (Ames et al., 1993; Doll, 1990; Dragsted et al., 1993; Willett, 1994a). The antitumorigenic effects of vegetables were also found in experiments using cells (Maeda et al., 1992) and animals (Belman, 1983; Bingham, 1990; Bresnick et al., 1990; Maltzman et al., 1989; Stoewsand et al., 1988; Stoewsand et al., 1989; Wattenberg and Coccia, 1991). There is a highly significant negative association between intake of total fruits and vegetables and cardioand cerebrovascular disease mortality (Acheson and Williams, 1983; Armstrong et al., 1975; Burr and Sweetnam, 1982; Phillips et al., 1978; Verlangieri et al., 1985). Vegetarians and nonvegetarians with a high intake of fruits and vegetables also have reduced blood pressure (Ascherio et al., 1992; Sacks and Kass, 1988). The protection that fruits and vegetables provide against diseases, including cancer and cardio- and cerebrovascular diseases, has been attributed to the various antioxidants, especially antioxidant vitamins, including ascorbic acid and R-tocopherol, contained in these fruits and vegetables (Ames, 1983; Gey, 1990; Gey et al., 1991; Riemersma et al., 1989; Sta¨helin et al., 1991a,b; Steinberg et al., 1989, 1991; Willett, 1994b). However, the majority of the antioxidant activity of a fruit or vegetable may be from compounds other than vitamin C, vitamin E, or β-carotene. For example, some flavonoids that are often found in the human diet have antioxidant activities (Bors and Saran, 1987; Bors et al., 1990; Hanasaki et al., 1994). antioxidant activities which cannot be accounted for by their vitamin C content (Wang et al., 1996). We also found that some flavonoids had much stronger antioxidant activities against peroxyl radicals than vitamin E, vitamin C, and glutathione (Cao et al., in press). The objective of this study was to determine the antioxidant capacities of 22 common vegetables, one green tea, and one black tea by using the oxygen radical absorbance capacity (ORAC) assay (Cao et al., 1993, 1995). Three different reactive species were used in the ORAC assay: (i) 2,2′-azobis(2-amidinopropane) dihydrochloride (AAPH), a peroxyl radical (ROO•) generator, (ii) Cu2+H2O2, mainly a hydroxyl radical (OH•) generator, and (iii) Cu2+, a transition metal. MATERIALS AND METHODS Chemicals. β-Phycoerythrin (β-PE) from Porphydium cruentum was purchased from Sigma (St. Louis, MO). The β-PE that was used in these experiments usually lost more than 90% of its fluorescence within 30 min in the presence of 4 mmol/L AAPH. AAPH was purchased from Wako Chemicals USA Inc. (Richmond, VA). 6-Hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid (Trolox) was obtained from Aldrich (Milwaukee, WI). Tea and Vegetables. Twenty-two vegetables were purchased on three separate occasions from local supermarkets. The 22 vegetables were garlic, kale, spinach, Brussels sprouts, alfalfa sprouts, broccoli flowers, beets, red bell pepper, onion, corn, eggplant, cauliflower, potato, sweet potato, cabbage, leaf lettuce, string bean, carrot, yellow squash, iceberg lettuce, celery, and cucumber. The green tea used in the study was Chin Chu oriental blend tea. The black tea (all black teas are fermented teas) was a dried powder and provided by Tea Trade Health Research Association. Sample Preparation. The black tea was completely dissolved in deionized water (5 mg/mL) and used for ORAC assay directly after suitable dilution with phosphate buffer (75 mM, pH 7.0). The green tea was brewed for 30 min in deionized water (1:60, w/v, 95-100 °C). The edible portion of a vegetable was weighed and then homogenized by using a blender after adding deionized water (1:2, w/v). The brewed green tea and vegetable homogenate were then centrifuged

at 34000g for 30 min (4 °C). The supernatant (water soluble fraction) was recovered and used directly for the ORAC assay after suitable dilution with the phosphate buffer. The pulp (water insoluble fraction) was washed twice with deionized water and further extracted by using pure acetone (1:4, w/v) with shaking at room temperature for 30 min. Acetone has been used by our laboratory (Wang et al., 1996) and others (Daniel et al., 1989; Mass et al., 1991) to extract antioxidants from fruit pulp. The acetone extract was recovered after centrifugation (34000g, 10 min, 4 °C), and the sample was used for the ORAC assay after suitable dilution with phosphate buffer. The ORAC activity of a vegetable or the green tea was calculated by adding the ORAC activity from its water soluble fraction and its pulp fraction extracted with acetone. The dry matter of a vegetable was determined after drying the vegetable at 40 °C for 1 week. Automated ORAC Assay. The automated ORAC assay was carried out on a COBAS FARA II spectrofluorometric centrifugal analyzer (Roche Diagnostic System Inc., Branchburg, NJ) with fluorescent filters (ex, 540 nm; em, 565 nm). The procedure was based on a previous report of Cao and coworkers (Cao et al., 1993), as modified for the COBAS FARA II (Cao et al., 1995). Briefly, in the final assay mixture (0.4 mL total volume), β-PE (16.7 nM) was used as a target of free radical (or oxidant) attack, with either (i) AAPH (4 mM) as a peroxyl radical generator (ORACROO• assay), (ii) H2O2-Cu2+ [H2O2, 0.3%; Cu2+ (as CuSO4), 9 µM] as mainly a hydroxy radical generator (ORACOH• assay), or (iii) Cu2+ (as CuSO4) (18 µM) as a transition metal oxidant (ORACCu assay). Trolox was used as a control standard. A 0.1 mM stock solution was stable for at least 1 month at -80 °C. The analyzer was programmed to record the fluorescence of β-PE every 2 min after AAPH, H2O2-Cu2+, or Cu2+ was added. All fluorescent measurements were expressed relative to the initial reading. Final results were calculated using the differences of areas under the β-PE decay curves between the blank and a sample and are expressed as µmol of Trolox equiv/g of tea or vegetables (Cao et al., 1993, 1995), except when Cu2+ alone (i.e., without H2O2) was used as an oxidant in the assay. In the presence of Cu2+ alone, Trolox cannot be used as an antioxidant standard since Trolox may act as a prooxidant in the presence of Cu2+ (Cao and Cutler, 1993). Therefore, the result of the ORACCu assay in this case was calculated using (areasample - areablank)/areablank and expressed as antioxidant units; 1 unit equals the antioxidant activity which increases the area under the β-PE decay
curve by 100% in the ORACCu assay. A negative ORACCu value indicated a Cu2+-initiated prooxidant activity. RESULTS
The antioxidant activities against peroxyl radicals (ORACROO• activity) of 22 common vegetables, one green tea, and one black tea are shown in Table 1. Based on the fresh or wet weight of a vegetable, garlic and kale were in the top quintile of ORACROO• measured in the 22 vegetables. Spinach, Brussels sprouts, alfalfa sprouts, broccoli flowers, beets, red bell pepper, onion, corn, and eggplant had ORACROO• values that fell in the middle three quintiles (3.9-12.6). Cauliflower, potato, sweet potato, cabbage, leaf lettuce, string beans, carrot, yellow squash, iceberg lettuce, celery, and cucumber were in the lowest quintile of ORACROO• activities of the vegetables measured. However, based on the dry weight of a vegetable, kale had the highest ORACROO• activity followed by spinach, alfalfa sprouts, beets, eggplant, red bell pepper, Brussels sprouts, broccoli flowers, leaf lettuce, garlic, cauliflower, onion, and iceberg lettuce. Cabbage, string beans, carrots, corn, yellow squash, cucumber, potato, sweet potato, and celery were in the lowest quintile (below 32.0) of ORACROO• activities expressed on a dry matter basis. Green and black teas had much higher ORACROO• activities than any of the vegetables studied (4.5-5-fold higher than kale and 6070-fold higher than celery, based on the dry weight). The antioxidant activities against hydroxyl radicals (ORACOH• activity) of the vegetables and green tea are also shown in Table 1. Based on the fresh or wet weight of a vegetable, kale had the highest ORACOH• activity followed by Brussels sprouts, alfalfa sprouts, beets, spinach, broccoli flowers, corn, string beans, cabbage, leaf lettuce, eggplant, cauliflower, yellow squash, garlic, potato, sweet potato, carrot, iceberg lettuce, red bell pepper, onion, celery, and cucumber. garlic, and broccoli flowers, all act as antioxidants against Cu2+, and their antioxidant activity increased as their concentration increased in the assay system. Figure 2 presents the calculated ORACROO• intake based upon a common measured size or serving. For many of the vegetables this common measured proportion represents a 1/2 cup serving size except for garlic (1 clove), onion (1 tablespoon), potato (1 potato), and lettuce (1 leaf). In Figure 2, the common serving size is presented in grams. Based upon this calculation, kale, beets, red bell pepper, Brussels sprouts, broccoli flowers, spinach, potatoes, and corn likely provide a significant amount of ORACROO• in the diet if these vegetables are consumed on a regular basis. Frequency of consumption of the individual vegetables would be the other factor determining which vegetables contribute the most to the ORAC consumed in a common diet. DISCUSSION

lettuce, eggplant, cabbage, broccoli flower, cauliflower, yellow squash, corn, carrot, cucumber, red bell pepper, celery, potato, sweet potato, onion, and garlic. The ORACOH• activity of green tea, based on dry weight, was between that of beets and spinach. Green tea showed a prooxidant activity (negative ORACCu activity) in the presence of Cu2+ (without H2O2) (Table 1). This Cu2+-initiated prooxidant activity, however, was not found in any vegetables evaluated in this study. Based on the fresh or wet weight of the vegetable, garlic had the highest antioxidant activities against Cu2+ (ORACCu activity) followed by broccoli flowers, spinach, and the others. However, spinach had the highest ORACCu activity, if activity was based on the dry weight, followed by iceberg lettuce, broccoli flowers, and the others. The ‘antioxidant score’ of a vegetable shown in Table 1 was calculated by simply adding ORACROO• (µmol of Trolox equiv), ORACOH• (µmol of Trolox equiv), and ORACCu (103 units), based on the wet weight of the vegetable. One nanomole of Trolox equivalent calculated from ORACROO• assay and 1 ORACCu unit calculated from ORACCu assay represent a similar area difference under the β-PE decay curve between the blank and a sample, which was used in the ORAC quantification. Because ORACROO• activity of a vegetable weights the score more heavily than ORACOH• or ORACCu activity of the vegetable in the scoring system, the ‘antioxidant score’ did not rank the vegetables in a significantly different order than what was observed with the ORACROO• assay. The ‘antioxidant score’ was not given for the teas since they are dry, not fresh. The ORACCu activities of tea and vegetables were determined using different extract concentrations, since the Cu2+-initiated prooxidant activity of some antioxidants is seen only at a relatively high concentration (Cao and Cutler, 1993). The results in Figure 1 show that in the presence of Cu2+ (without H2O2), tea acts as a prooxidant at all concentrations, and the prooxidant activity increased with increased tea concentration. However, of the tested vegetables including spinach,
The ORAC assay developed recently by Cao and coworkers (Cao et al., 1993, 1995) provides a unique and novel way to evaluate the potential antioxidant activities of various compounds and biological samples. This method is superior to other similar methods for two reasons. First, the ORAC assay system uses an areaunder-curve (AUC) technique and thus combines both inhibition time and inhibition degree of free radical action by an antioxidant into a single quantity (Cao et al., 1995). Other similar methods (Ghiselli et al., 1994; Glazer, 1990; Miller et al., 1993; Wayner et al., 1985; Whitehead et al., 1992) use either the inhibition time at a fixed inhibition degree or the inhibition degree at a fixed time as the basis for quantitating the results. Second, different free radical generators or oxidants can be used in the ORAC assay. This is important because the measured antioxidant activity of a biological sample depends upon which free radical or oxidant is used in the assay (Cao et al., 1996a,b). Peroxyl radical (ROO•) is a common free radical found in the body and used in the antioxidant activity assays (Wayner et al., 1985; Glazer, 1990; Cao et al., 1993, 1995; Ghiselli et al., 1994). It is slightly less reactive than OH• and thus possesses an "extended " half-life of seconds instead of nanoseconds (Grisham, 1992). The total antioxidant capacity of some common fruits was thus determined by us using the ORACROO• assay (Wang et al., 1996), which measures all traditional antioxidants including ascorbic acid, R-tocopherol, β-carotene, glutathione, bilirubin, uric acid, melatonin (Cao et al., 1993; Pieri et al., 1994), and flavonoids (Cao et al., in press). In the current study, Cu2+-H2O2 (a “OH•” generator) and Cu2+ alone were also used to assess the antioxidant activities of one green tea and 22 vegetables. Most of the “OH•” thought to be generated in vivo comes from metal-dependent reduction of H2O2, except during abnormal exposure to ionizing radiation. In vitro the metal can be titanium, copper, iron, or cobalt, but the best candidates for promoters of OH• formation in vivo seem to be iron and, to a smaller extent, copper. Cu2+H2O2 or Cu2+ alone is frequently used in inducing oxidative damage to protein and nucleic acids (Parthasarathy et al., 1989; Sato et al., 1992). The ORACOH• assay with Cu2+-H2O2 as a OH• generator measures compounds like mannitol, glucose, uric acid (at physiological concentrations), proteins, and transition metal chelators, but not compounds, such as ascorbic acid, that react directly with copper and produce reactive species.

The ORACCu assay using copper alone measures not only the antioxidant activity (positive ORACCu value) of a compound which can sequester transition metals but also the transition metal-initiated prooxidant activity (negative ORACCu value) of a compound, such as ascorbic acid (Cao and Cutler, 1993) and some flavonoids (Cao et al., in press). At this point, we do not have a good indication as to which radical generator provides the ‘best’ estimate of antioxidant activity of the vegetables. Perhaps, the formulation of an ‘antioxidant score’, which takes into account the antioxidant activities determined by the three different reactive species or oxidants, can give us some additional useful information. We calculated the ‘antioxidant score’ of each vegetable in this study by simply adding ORACROO•, ORACOH•, and ORACCu values, based on wet weight of the vegetable, since 1 nmol of Trolox equiv calculated from ORACROO• assay and 1 ORACCu unit calculated from ORACCu assay represent a similar area difference under the β-PE decay curve between the blank and a sample. The ORACROO• value of a vegetable weights the score more heavily than the ORACOH• or ORACCu value of the vegetable in the scoring system, which also seems reasonable because peroxyl radicals tend to be more prevalent in biological systems. However, the ‘antioxidant score’ did not rank the vegetables in a significantly different order than what was observed with the ORACROO• assay. Our results demonstrated clearly that all vegetables tested in this study had antioxidant activities against not only peroxyl radicals but also hydroxyl radicals and transition metals (Cu2+), although their ORACROO•, ORACOH•, and ORACCu activities vary considerably from one kind of vegetable to another. It is an important finding that these vegetables (and also fruits like strawberry, unpublished data) acted as antioxidants when a transition metal oxidant was used in the ORAC assay and the antioxidant activity also increased as their concentrations increased. The transition metalinitiated prooxidant actions of ascorbic acid (Beach and
Giroux, 1992) and R-tocopherol (Iwatsuki et al., 1995; Maiorino et al., 1993; Yoshida et al., 1994) have been described. Using Cu2+-H2O2 in the ORAC assay, it was also found that ascorbic acid and Trolox (at a relatively high concentration), a water soluble R-tocopherol analogue, acted as prooxidants (Cao and Cutler, 1993). Therefore, in terms of the antioxidant quality in vitro, the natural antioxidant mixture contained in fruits or vegetables appears to be better than a single antioxidant or a simple antioxidant mixture of ascorbic acid, R-tocopherol, and β-carotene. The antioxidant capacity varies considerably from one kind of vegetable to another, similar to what we found in fruits (Wang et al., 1996). The ORACROO• activities of kale and spinach were similar to that observed in strawberries (Wang et al., 1996) whether the data were based on wet or dry weight. For example, based on the wet weight of a fresh vegetable, the ORACROO• activity (which measures all traditional antioxidants) for kale was about 2 times the activity measured in beet and broccoli flowers, 8-9 times the activity measured in carrots and string beans, and 29-35 times the activity measured in celery and cucumber. Based upon a common measured or serving size, kale, beets, red peppers, Brussels sprouts, broccoli flowers, spinach, potatoes, and corn likely provide the largest amount of ORACROO• consumed from vegetables (Figure 2), although frequency of consumption of the individual vegetables would be another factor determining which vegetables contribute the most to ORAC consumed in a common diet. The green and black teas had much higher antioxidant activities against peroxyl radicals than all fruits and vegetables that we have examined. Their ORACROO• activity, based on dry weight, was 4.5-6.0 times the activity measured in kale and strawberry (Wang et al., 1996). The ORACOH• activity of the green tea, based on the dry weight, was only 58% of that measured in kale. The ORACOH• activity of the tea was actually compromised by the Cu2+ used in the assay, since the

ORACCu activity of the tea was negative, indicating a prooxidant activity of the tea in the presence of Cu2+. It seems clear that some tea components can absorb hydroxyl radicals and other reactive species produced from the reaction between Cu2+ and H2O2. Some tea components apparently can produce reactive species through direct reactions among these tea components, Cu2+, and O2, when Cu2+-H2O2 is used as a reactive species generator in the ORAC assay. It is also possible that some tea components, such as some flavonoids (Cao et al., in press), can play these two opposite roles at the same time. However, the transition metal-initiated prooxidant actions of tea, ascorbic acid, and R-tocopherol may not be important in vivo, where transition metals will be largely sequestered, except perhaps in certain diseases involving metal overload. Recent experiments have already demonstrated the inhibition by tea and tea polyphenols of tumorigenesis in different animal models (Yang and Wang, 1993), although the effect of tea consumption on cancer risk in humans as revealed by epidemiologic studies is less clear (International Agency for Research on Cancer, 1991). The antioxidant defense system of the body is composed of different antioxidant components. The antioxidant capacities of these antioxidant components depend upon which free radicals or oxidants are produced in the body. Some fruits and vegetables contain a group of natural antioxidants that have not only a high antioxidant activity but also a good antioxidant quality. Therefore, the supplementation of these natural antioxidants through a balanced diet containing enough fruits and vegetables could be much more effective and economical than the supplementation of an individual antioxidant, such as ascorbic acid or R-tocopherol, in protecting the body against various oxidative stresses. In summary, the antioxidant activities of 22 common vegetables, one green tea, and one black tea were measured using the automated ORAC assay with three different reactive species: a peroxyl radical generator, a hydroxyl radical generator, and Cu2+, a transition metal. Based on the fresh weight of a vegetable, garlic had the highest antioxidant activity against peroxyl radicals followed by kale, spinach, Brussels sprouts, alfalfa sprouts, and others, while kale had the highest antioxidant activity against hydroxyl radicals followed by Brussels sprouts, alfalfa sprouts, beets, spinach, and the others. Kale also had the highest ORACROO• activity, when results were expressed on a dry weight basis. The green and black teas had much higher antioxidant activity against peroxyl radicals than all of the vegetables tested in this study. However, the tea exhibited a prooxidant activity in the presence of Cu2+, which has also been reported for the antioxidants, ascorbic acid and R-tocopherol; this prooxidant activity was not found in the vegetables analyzed in this study. Therefore, the supplementation of natural antioxidants through a balanced diet containing enough fruits and vegetables could be the most effective in protecting the body against various oxidative stressors. Tế bào da thường xuyên tiếp xúc với các loại oxy hoạt động (ROS) vàstress oxy hóa(oxidative stress) từ các nguồn ngoại sinh và nội sinh. Bức xạ UV là yếu tố môi trường quan trọng nhất trong sự phát triển của ung thư da và lão hóa da. Bức xạ cực tím (Ultraviolet radiation - UVR) là yếu tố nguy cơ cho sự phát triển của tổn thương da sớm cũng như ung thư tế bào hắc tố (melanoma) haykhông phải tế bào hắc tố (nomelanoma).Ung thư da thường phát triển ở lớp biểu bì (lớp ngoài cùng của da), do đó khối u có thể thấy rõ ràng, dễ dàng phát hiện.Có nhiều loại ung thư da. Một trong số đó hình thành từ tế bào hắc tố da – melanoma. Các loại khác bao gồm các ung thư của biểu mô tế bào như ung thư biểu mô tế bào đáy và tế bào vảy. Các loại ung thư biểu mô này (ung thư biểu mô tế bào đáy và tế bào vảy) còn được gọi chung là “Ung thư không do tế bào hắc tố” (nonmelanoma skin cancers).

Trong khi phơi nhiễm với tia UVR là yếu tố nguy cơ gắn liền với sự phát triển của ung thư da, các yếu tố môi trường khác(như bức xạ ion hóa, ngộ độc arsenicmãn tính và ức chế miễn dịch) và các yếu tố di truyền (như tiền sử gia đình, loại da và hệ thống di truyền) cũng có thể góp phần gây ung thư. Tiếp xúc UVR thúc đẩy hình thành ung thư qua hai cơ chế: thứ nhất là sự đột biến của DNA tế bào biểu bì, làm khởi phát khối u.Thứ hai là kết hợp với quá trình ức chế miễn dịch, làm cho khối u phát triển dần dần mà không bị sự giám sát của hệ thống miễn dịch[1].

ROS. Các chất nội sinh bảo vệ da là melanin và các enzym chống oxy hóa. Melanin (chất được tạo ra bởi các tế bào biểu bì hắc tố - melanocytes)là yếu tố đầu tiên phòng ngừa sự tổn hại DNA ở bề mặt da nhưng nó không thể ngăn ngừa hoàn toàn được các tổn thương da. Cách phòng ngừa thứ hai là quá trình sửa chữa, tức là loại bỏ các phân tử sinh học bị hư hỏng trước khi chúng có thể tích tụ và sự hiện diện của chúng làm thay đổi sự chuyển hóa của tế bào.Bảo vệ khỏi tia UV bao gồm tránh ánh nắng mặt trời, sử dụng kem chống nắng, quần áo bảo vệ và các chất bổ sung chống oxy hoá.

Bức xạ mặt trời đã được nghiên cứu về khả năng gây ung thư trong nhiều nghiên cứu trên chuột nhắt và chuột cống.Một số lượng lớn động vật được nghiên cứu, và các khối u lành tính và ác tính trên da phát triển trong hầu hết các động vật sống sót. Mặc dù các nghiên cứu thiếu các chi tiết định lượng, kết quả cho thấy bằng chứng thuyết phục rằng ánh sáng mặt trời có khả năng gây ung thư cho da động vật[3]. Mặt dù thương tổn DNA do hình thành ROS không phải ít xẩy ra bởi vì người ta ước tính rằng tế bào người trung bình có 105 lần oxy hóa mỗi ngày do quá trình chuyển hóaoxy hóa tế bào[4]. DNA có chức năng rất ổn định, do đó tỷ lệ mắc ung thư thấp hơn dự tính, kể cả tần số xuất hiện oxy hóa cao. DNA.Hầu hết sự phá vỡ DNA được sửa chữa bởi các protein hiện diện trong nhân tế bào, nhưng những tổn thương di truyền không được sửa chữa của DNA có thể dẫn đến ung thư da. Các cơ chế quan trọng ngăn ngừa tác hại của oxy hóa bao gồm các enzym chống oxy hóa và hệ thống bảo vệnonenzym như các quá trình sữa chữa. Nhưng khi tuổi tác tăng lên, các cơ chế chống oxy hóa nội sinh và quá trình sữa chữa không làm việc hiệu quả nữa. Việc xem các phản ứng gốc tự do như là sự khởi xướng và thúc đẩy quá trình sinh ung thư ngụ ý rằng những can thiệp nhằm hạn chế hoặc ức chế các yếu tố này có thể làm giảm nguy cơ mắc ung thư. Chất chống oxy hóa sinh học được định nghĩa là bất kỳ chất nào có nồng độ thấp so với chất nền có khả năng làm giảm hoặc ngăn ngừa sự oxy hóa của chất nền đó[6]. Các khả năng chống oxy hóa bao gồm giảm stress oxy hóa, tổn thương DNA, chuyển hóa ác tính, và các chỉ số tổn thương tế bào trên in vitro cũng như trong dịch tễ học vớitỷ lệ thấp hơn các loại ung thư và bệnh lão hóa.Các chất chống oxy hóa làm giảm các tác hại của ROS và có thể làm suy yếu và/hoặc đảo ngược các quá trình có thể sinh ra độc tính hoặc bệnh tật. Song, sự gia tăng và kéo dài hoạt động gốc tự do có thểlấn át cơ chế bảo vệ ROS, góp phần gia tăng các bệnh về da, rối loạn và ung thư da. Tất cả các enzym chống oxy hóa đều có trong da nhưng vai trò của chúng trong việc bảo vệ tế bào chống lại các tác hại oxy hóa do UVR chưa được làm sáng tỏ. Để chống lại sự tấn công của ROS, da đã phát triển một hệ thống chống oxy hóa phức tạp trong đó có manganese-superoxide dismutase (MnSOD). Nghiên cứu của Poswig và cộng sự[13] cho thấy rằng đáp ứng phản ứng chống oxy hóa của MnSOD có thể xẩy ra khi chiếu tia UVA lặp đi lặp lại. Tác giả cung cấp bằng chứng cho sự tăng đáp ứng của MnSOD trên sự chiếu xạ UVA lặp đi lặp lại có thể góp phần vào đáp ứng hiệu quả UVA của da trong quá trình điều trị quang trị liệu xơ cứng da. 4. Vitamin C

Bổ sung vitamin C đường uống (500mg / ngày) được thực hiện bởi 12 tình nguyện viên trong 8 tuần, dẫn đến tăng đáng kể hàm lượng vitamin C trong huyết tương và da[15]. Sự bổ sung không ảnh hưởng đến UVR gây ra phản ứng ban đỏ. Nồng độ malonaldehyd ở da giảm đi do bổ sung vitamin C, nhưng ngạc nhiên là sự giảm hàm lượng ở da của các glutathione và protein thiols. Việc da tiếp xúc một mình hay kết hợp tia UV và Ozon tạo ra sự suy giảm vitamin E đáng kể[19], điều này có nghĩa là vitamin E chống lại các ROS một cách hiệu quả khi da tiếp xúc với UVR. Sự suy giảm vitamin E là một trong những dấu hiệu stress oxy hóa sớm nhất khi da người tiếp xúc với UVR hay các môi trường nguy cơ khác[20]. 6.ߚ ߚ-Caroten
ߚ-carotene là thành phần chủ yếu của nhiều sản phẩm sẵn có trên thị trường góp phần vào hệ thống quang bảo vệ của da. Bổ sung ߚ-carotene thường dùng như là các chất bảo vệ ánh nắng mặt trời đường uống, nhưng các nghiên cứu về hiệu quả tác dụng chống lại các phản ứng của da với ánh nắng mặt trời của ߚ-carotene đường uống rất ít và các kết quả còn mâu thuẫn[23]. Các nghiên cứu về sử dụng ߚ-carotene cho thấy rằng sử dụng liều 15-30mg/ngày trong 10-12 tuần có tác dụng bảo vệ tia UV gây ra ban đỏ. Một thử nghiệm ngẫu nhiên, nghiên cứu lâm sàng có đối chứng hiệu quả của ߚ-carotene đường uống (50mg/ngày trong 5 năm) ở bệnh nhân dự phòng ung thư da với bệnh nhân ung thư da nonmelanoma gần đây cho thấy tác dụng không rõ ràng của ߚ-carotene ở số lượng lẫn hoặc thời gian xuất hiện ung thư da[27]. Các phân tử AND liên tục bị “bắn phá” bởi ROS có nguồn gốc từ các quá trình nội sinh cũng như các tác nhân môi trường bên ngoài và các nguồn bức xạ. Các chất chống oxy hóa có thể hoạt động bằng cách trung hòa các gốc tự do và bằng cách tăng cường hệ thống sữa chữa enzym DNA thông qua sự điều khiển gen sau phiên mã của các nhân tố phiên mã[41]. Khả năng sữa chữa của thế bào da người có liên quan trực tiếp đến xác suất khởi phát quá trình sản sinh ung thư và cuối cùng là hình thành khối u. Ngày càng nhiều bằng chứng cho thấy rằng thay đổi chế độ ăn và chế độ dinh dưỡng có thể làm giảm stress oxy hóa, sự hình thành gốc tự do và do đó làm chậm các quá trình tổn thương da. Sử dụng các chất chống oxy hóa ở da sau khi bị tổn thương do UVR có thể gây thêm các tác dụng có hại trong kiểm soát chu trình tế bào và quá trình tế bào chết theo chương trình. Skin cells are constantly exposed to reactive oxygen species (ROS) and oxidative stress from exogenous and endogenous sources. UV radiation is the most important environmental factor in the development of skin cancer and skin aging. The primary products caused by UV exposure are generally direct DNA oxidation or generation of free radicals which form and decompose extremely quickly but can produce effects that can last for hours, days, or even years. UV-induced generation of ROS in the skin develops oxidative stress when their formation exceeds the antioxidant defense ability. The reduction of oxidative stress can be achieved on two levels: by lowering exposure to UVR and/or by increasing levels of antioxidant defense in order to scavenge ROS. The only endogenous protection of our skin is melanin and enzymatic antioxidants. Melanin, the pigment deposited by melanocytes, is the first line of defense against DNA damage at the surface of the skin, but it cannot totally prevent skin damage. A second category of defense is repair processes, which remove the damaged biomolecules before they can accumulate and before their presence results in altered cell metabolism. Additional UV protection includes avoidance of sun exposure, usage of sunscreens, protective clothes, and antioxidant supplements.
1. Introduction Ultraviolet radiation (UVR) is an essential risk factor for the development of premalignant skin lesions as well as of melanoma and nonmelanoma skin cancer. Skin cancer generally develops in the epidermis (the outermost layer of skin), so a tumor is usually clearly visible, which makes it easier to detect. There are various types of skin cancer. One main class is formed by the cutaneous melanocytes—melanoma. The other main types are basal cell carcinoma and squamous cell carcinoma, cancers of the epithelial cells. These carcinomas of the skin (basal cell and squamous cell carcinomas) are sometimes, collectively, called “nonmelanoma skin cancers.” While exposure to UVR is the risk factor most closely linked to the development of skin cancer, other environmental factors (such as ionizing radiation, chronic arsenic ingestion, and immunosuppression) and genetic factors (such as
family history, skin type, and genetic syndromes) also potentially contribute to carcinogenesis. UVR exposure appears to promote the induction of skin cancer by two mechanisms. The first involves direct mutagenesis of epidermal DNA, which promotes the induction of neoplasia. The second is associated with immune suppression, which allows the developing tumor to escape immune surveillance and grow progressively [1]. It is known that UVR exposure results in photochemical modification of the genetic material (DNA), but most of this damage is accurately and efficiently repaired by the cell. However, if the amount of damage is too great, some of the alterations to the DNA may remain as permanent mutations. DNA absorbs UV light, and the absorbed energy can break bonds in the DNA. Most of the DNA breakages are repaired by proteins present in the cell nucleus, but unrepaired genetic damage of the DNA can lead to skin
2 cancers. As already mentioned previously, solar UVR induces a variety of photoproducts in DNA, including cyclobutanetype pyrimidine dimers, pyrimidine-pyrimidone (6–4) photoproducts, thymine glycols, cytosine damage, purine damage, DNA strand breaks, and DNA-protein crosslinks. It has been proposed that if unrepaired damage occurs to regulatory genes (e.g., tumor suppressor genes), this may be involved in the process of carcinogenesis. In this context, mutations to and activation of genes may be important. Other responses likely to result from UVR exposure of cells include increased cellular proliferation, which could have a tumor-promoting effect on genetically altered cells, as well as changes in components of the immune system present in the skin [2]. Solar radiation was tested for carcinogenicity in a series of exceptional studies in mice and rats. Large numbers of animals were studied, and well-characterized benign and malignant skin tumors developed in most of the surviving animals. Although the reports are deficient in quantitative details, the results provide convincing evidence that sunlight is carcinogenic for the skin of animals [3]. Although DNA damage due to reactive oxygen species formation is not a rare event since it is estimated that human cell sustains an average of 105 oxidative hits per day due to cellular oxidative metabolism [4], DNA is functionally very stable, so that the incidence of cancer is much lower than one would expect, taking into account the high frequency of oxidative hits. It seems that in rapidly dividing epithelium, such as the epidermis, nuclear damage triggered by some xenobiotics may not be so important because of the constant introduction of new healthy cells, whereas a DNA mutation has a much higher probability to become fixed to a transformed phenotype in tissues (e.g., liver) with slow cell turnover [5]. This may explain at least in part why the absolute number of clinically well-recognizedhuman skin carcinogens is so small. The specific mutations needed to activate an oncogene would be rarer. The probability of mutating five genes needed for cancer formation, such as an oncogene and both alleles of two particular tumor suppressor genes, is at best 10−20 . With 106 proliferating keratinocytes per cm2 in human skin, and âˆź1 cm2 exposed, less than one person in 1010 would develop a tumor. However, clonal expansion increases by 1,000-fold the number of targets for the next mutation and increases the probability of tumor formation. It is widely believed that cancer development inhumans and laboratory animals is caused by sequential mutations and clonal outgrowth of somatic cells. Most important oxidative damage prevention mechanisms include antioxidative enzymatic and nonenzymatic defenses as well as repair processes. But the problem arises with age, when endogenous antioxidative mechanisms and repair processes do not work anymore in the effective way. The identification of free radical reactions as initiators and promoters of the cancer process implies that interventions aimed at limiting or inhibiting these factors should be able to reduce the rate of cancer incidence. There still remains the answer regarding controversial data on the use of synthetic antioxidants in cancer prevention and treatment.

2. Skin Antioxidant Defenses Although the skin possesses an elaborate antioxidant defense system to deal with oxidative stress, excessive and chronic exposure to UV light or other oxidizing agents (e.g., cigarette smoke) can overwhelm the cutaneous antioxidant and immune response capacity, leading to oxidative damage and immunotoxicity, premature skin aging, and skin cancer. A biological antioxidant has been defined as any substance that when present at low concentrations compared to those of an oxidizable substrate significantly delays or prevents oxidation of that substrate [6]. Antioxidant functions are associated with lowering oxidative stress, DNA damage, malignant transformation, and other parameters of cell damage in vitro as well as epidemiologically with lowered incidence of certain types of cancer and degenerative diseases. Antioxidants attenuate the damaging effects of reactive oxygen species (ROS) and can impair and/or reverse many of the events that contribute to epidermal toxicity and disease. However, increased or prolonged free radical action can overwhelm ROS defense mechanisms, contributing to the development of cutaneous diseases, disorders, and skin cancer. The two main categories of antioxidant defenses are those whose role is to prevent the generation of ROS and those that intercept any radicals that are generated [7]. The defense system exists in aqueous and membrane compartments of cells and can be enzymatic and nonenzymatic. A second category of natural antioxidants are repair processes, which remove the damaged biomolecules before they accumulate to cause altered cell metabolism or viability [7]. The skin is equipped with a network of protective antioxidants. They include enzymatic antioxidants such as glutathione peroxidase, superoxide dismutase and catalase, and nonenzymatic low-molecular-weight antioxidants such as vitamin E isoforms, vitamin C, glutathione (GSH), uric acid, and ubiquinol [8]. Various other components present in skin are potent antioxidants including ascorbate, uric acid, carotenoids, and sulfhydrils. Water-soluble antioxidants in plasma include glucose, pyruvate, uric acid, ascorbic acid, bilirubin, and glutathione. Lipid-soluble antioxidants include alpha-tocopherol, ubiquinol-10, lycopene, �-carotene, lutein, zeaxanthin, and alpha-carotene. In general, the outer part of the skin, the epidermis, contains higher concentrations of antioxidants than the dermis [9, 10]. In the lipophilic phase, �-tocopherol is the most prominent antioxidant, while vitamin C and GSH have the highest abundance in the cytosol. On molar basis, hydrophilic nonenzymatic antioxidants including L-ascorbic acid, GSH, and uric acid appear to be the predominant antioxidants in human skin [11]. Their overall dermal and epidermal concentration are more than 10- to 100-fold greater than those found for vitamin E or ubiquinol. The antioxidant capacity of the human epidermis is far greater than that of dermis. This was demonstrated in the studies by Shindo et al. [9, 10] where enzymatic and nonenzymatic antioxidants in human epidermis and dermis from six healthy volunteers undergoing surgical procedures were measured. A similar study was done by Shindo et al. [8] where enzymatic and nonenzymatic antioxidants in
Oxidative Medicine and Cellular Longevity epidermis and dermis of hairless mice were compared. Catalase, glutathione peroxidase, and glutathione reductase were higher in epidermis than dermis. Lipophilic antioxidants (alpha-tocopherol, ubiquinol 9, and ubiquinone 9) and hydrophilic antioxidants (ascorbic acid, dehydroascorbic acid, and glutathione) were also higher in epidermis than in dermis. The stratum corneum (SC) was found to contain both hydrophilic and lipophilic antioxidants. Vitamins C and E (both �- and �-tocopherols) as well as GSH and uric acid were found to be present in the SC [12]. Surprisingly, they were not distributed evenly, but in gradient fashion, with low concentrations on the outer layers and increasing concentrations toward the deeper layers of the SC. This phenomenon may be explained by the fact that O2 partial pressure is higher in the upper SC, which already causes a mild oxidative stress resulting in the partial depletion of antioxidants. Taken together, all the major antioxidant enzymes are present in skin but their role in protecting cells against oxidative damage generated by UVR has not been elucidated. In response to the attack of ROS, the skin has developed a complex antioxidant defense system including, among others, the manganese-superoxide dismutase (MnSOD). The study of Poswig et al. [13] revealed that adaptive antioxidant response of manganese-superoxide dismutase following repetitive UVA irradiation can be induced. The authors provide evidence for the increasing induction of MnSOD upon repetitive UVA irradiation that may contribute to the effective adaptive UVA response of the skin during light hardening in phototherapy. The study of Fuchs et al. [5] on mouse skin showed that acute UV exposures lead also to changes in glutathione reductase and catalase activity in mouse skin but to insignificant changes in superoxide dismutase and glutathione peroxidase. The study of Sander et al. [14] confirmed that chronic and acute photodamage are mediated by depleted antioxidant enzyme expression and increased oxidative protein modifications.
3. The Importance of Antioxidants in Decreasing ROS Formation and Skin Cancer Prevention The only protection of our skin against UVR is its endogenous protection (melanin and enzymatic antioxidants) and antioxidants we consumed with the food (vitamin A, C, E, etc.). Dietary antioxidants thus play a major role in maintaining the homeostasis of the oxidative balance. Vitamin C (ascorbic acid), vitamin E (�-tocopherol), beta-carotene, and other micronutrients such as carotenoids, polyphenols, and selenium have been evaluated as antioxidant constituents in the human diet. UVR exposure affects the skin antioxidants. Ascorbate, GSH, SOD, catalase, and ubiquinol are depleted in UV-B exposed skin, both dermis and epidermis. Levels of electron paramagnetic resonance (EPR) detectable ascorbyl radicals rise on UV exposure of skin. Studies of cultured skin cells and murine skin in vivo have indicated that UVRinduced damage involves the generation of ROS and depletion of endogenous antioxidant systems [15]. For example,
3 in the study by Shindo et al. [8], enzymatic and nonenzymatic antioxidants in epidermis and dermis and their responses to ultraviolet light of hairless mice were compared. After irradiation epidermal and dermal catalase and superoxide dismutase activities were greatly decreased. �-Tocopherol, ubiquinol 9, ubiquinone 9, ascorbic acid, dehydroascorbic acid, and reduced glutathione decreased in both epidermis and dermis by 26–93%. Oxidized glutathione showed a slight, nonsignificant increase. Many other studies confirmed that acute exposure of human skin to UVR in vivo leads to oxidation of cellular biomolecules that could be prevented by prior antioxidant treatment. There have been many studies performed where different antioxidants or combinations of antioxidants and different phytochemicals were tested in order to find evidence against ROS-induced damage. The outcomes of the studies examining the influence of exogenous antioxidants on the photoaging or damage protective effects, which are relevant for clinical practice, were in details presented elsewhere by Pandel et al. [16] and Poljsak et al. [17].
4. Vitamin C Oral vitamin C supplements (500 mg/day) were taken by 12 volunteers for 8 weeks resulting in significant rises in plasma and skin vitamin C content [15]. Supplementation had no effect on the UVR-induced erythemal response. The skin malonaldehyde content was reduced by vitamin C supplementation, but, surprisingly, reductions in the skin content of total glutathione and protein thiols were also seen. Authors speculate that this apparently paradoxical effect could be due to regulation of total reductant capacity by skin cells, such that vitamin C may have been replacing other reductants in these cells. Ascorbic acid was a photoprotectant in clinical human UV studies at doses just above the minimal erythema dose (MED). An opaque cream containing 5% ascorbic acid did not induce dermal sensitization in 103 human subjects. A product containing 10% ascorbic acid was nonirritant in a 4day minicumulative patch assay on human skin and a facial treatment containing 10% ascorbic acid was not a contact sensitizer in a maximization assay on 26 humans [15]. Many other studies have found that vitamin C can increase collagen production, protect against damage from UVA and UVB rays, correct pigmentation problems, and improve inflammatory skin conditions (reviewed in [16–18]).
5. Vitamin E Skin exposure to UV and ozone alone and in combination resulted in a significant potentiation of the UV-induced vitamin E depletion [19], which means that vitamin E is efficiently quenching ROS during UVR skin exposure. Depletion of vitamin E is one of the earliest oxidative stress markers in human skin exposed to UVR and other environmental stress [20]. One study showed that the number of sunburn to cells was decreased by treatment with the antioxidant tocopherol and may result from both direct protection from free radicals and indirect protection by means of increased epidermal
4 thickness [21]. Additionally, Packer and Valacchi [19] showed that vitamin E has skin barrier-stabilizing properties. Vitamin E provides protection against UV-induced skin photodamage through a combination of antioxidant and UV absorptive properties. Topical application of alpha-tocopherol on mouse skin inhibits the formation of cyclobutane pyrimidine photoproducts. However, topically applied alpha-tocopherol is rapidly depleted by UVB radiation in a dose-dependent manner [22].
6. �-Caroten �-carotene is a major constituent of commercially available products administered for systemic photoprotection. �carotene supplements are frequently used as so-called oral sun protectants, but studies proving a protective effect of oral treatment with �-carotene against skin responses to sun exposure are scarce and conflicting results have been reported [23]. Studies on the systemic use of �-carotene provide evidence that 15–30 mg/d over a period of about 10– 12 weeks produces a protective effect against UV-induced erythema. Similar effects have been attributed to mixtures of carotenoids or after long-term intake of dietary products rich in carotenoids. Supplementation with carotenoids contributes to basal protection of the skin but is not sufficient to obtain complete protection against severe UV irradiation [23]. Studies showed that the efficacy of �-carotene in systemic photoprotection depends on the duration of treatment and on the dose [23]. For successful intervention, treatment with carotenoids is needed for a period of at least ten weeks [24]. A study by Stahl et al. [25] was performed where carotenoids and tocopherols antioxidant effect was investigated against scavenging of ROS generated during photooxidative stress. It was investigated whether antioxidant oral supplementation may protect the skin from UV-induced erythema. The antioxidants used in this study provided protection against erythema in humans and may be useful for diminishing sensitivity to UV light. Heinrich et al. [26] additionally compared the erythema protective effect of betacarotene (24 mg/d from an algal source) to that of 24 mg/d of a carotenoid mix consisting of the three main dietary carotenoids, beta-carotene, lutein, and lycopene (8 mg/d each). A randomized, placebo-controlled clinical trial on the efficacy of oral �-carotene (50 mg/day over 5 years) in prevention of skin cancer in patients with recent nonmelanoma skin cancer showed no significant effect of �-carotene on either number or time of occurrences of new nonmelanoma skin cancer [27]. In a separate trial among healthy men, 12 years of supplementation with �-carotene (50 mg on alternate days) produced no reduction of the incidence of malignant neoplasms, including nonmelanoma skin cancer [28]. It must be pointed out that these intervention trials were conducted with patients whose skin cancer was primarily UV induced and it remains to be seen whether antioxidants are clinically effective in prevention of cutaneous chemocarcinogenesis [29]. Although the photoprotective effects of beta-carotene are thought to originate from its antioxidant properties, some studies documented prooxidant effects of beta-carotene.

7. Retinoids A study was done to compare the effects of dietary administration of a vitamin A drug (13-cis-retinoic acid) to the natural form of vitamin A (retinyl palmitate). Female mice were administered a chemical carcinogen to evaluate the incidence and severity on mouse skin tumor promotion. The results showed that retinyl palmitate inhibited the number and weight of tumors, whereas 13-cis-retinoic acid resulted in a decrease in weight but not in number of tumors promoted [30]. In another study, tumors were chemically induced in a group of Swiss mice over a 23-week period. The topical application of 13-cis-retinoic acid was compared to natural vitamin A (retinyl palmitate). This study showed that both retinyl palmitate and 13-cis-retinoic acid inhibited the development of skin papillomas and also had a marked effect on skin cancers [31].
8. Coenzyme Q10 It was recently reported that coenzyme Q10 protects against oxidative stress-induced cell death and enhances the synthesis of basement membrane components in dermal and epidermal cells [32]. Coenzyme Q10 (CoQ10) was reported to reduce ROS production and DNA damage triggered by UVA irradiation in human keratinocytes in vitro. Further, CoQ10 was shown to reduce UVA-induced MMPs in cultured human dermal fibroblasts [33]. It was reported that it is considered that CoQ10 appears to have also a cutaneous healing effects in vivo [34].
9. Glutathione In cell culture models using human skin cells, it has been clearly shown that glutathione depletion leads to a large sensitization to UVA (334 nm, 365 nm) and near-visible (405 nm) wavelengths as well as to radiation in the UVB (302 nm, 313 nm) [35, 36]. There is a direct correlation between the levels of sensitization and cellular glutathione content. Additional evidence that glutathione is a photoprotective agent in skin cells is derived from experiments which have demonstrated that glutathione levels in both dermis and epidermis are depleted by UVA treatment [37].
10. Green Tea In vitro and in vivo animal and human studies suggest that green tea polyphenols are photoprotective in nature and can be used as pharmacological agents for the prevention of solar UVB light-induced skin disorders including photoaging, melanoma, and nonmelanoma skin cancers after more clinical trials in humans. Topical treatment or oral consumption of green tea polyphenols (GTP) inhibits chemical carcinogenor UV radiation-induced skin carcinogenesis in different laboratory animal models. Topical application of GTP prior to exposure of UVB protects against UVB-induced local as well as systemic immune suppression in laboratory animals, which was associated with the inhibition of UVB-induced
Oxidative Medicine and Cellular Longevity infiltration of inflammatory leukocytes [38]. Another study of Vayalil et al. [39] demonstrated that topical application of green tea polyphenols reduced UVB-induced oxidation of lipids and proteins and depletion of antioxidant enzymes. Other protective effects include the reduced production of ROS and lipid peroxidation products, a reduced depletion of Langerhans cells and of endogenous antioxidant systems as reported by Afaq and Mukhtar [40].
11. Conclusions Skin DNA molecules are constantly “bombarded” by ROS originating from endogenous processes as well as from environmental agents and from radiation sources. Antioxidants might act by quenching free radicals and by enhancing the DNA enzyme repair systems through a posttranscriptional gene regulation of transcription factors [41]. The repair capacity of human skin cells therefore directly relates to the probability of initiation of the carcinogenesis process and eventually tumor formation. Evidence is accumulating that dietary changes and special nutrients may help to reduce oxidative stress and free radical formation and thereby slow down the skin damage process. Exogenous antioxidants like vitamins C and E and many others cannot be synthesized by the human body and must be taken up by the diet. Since the effectiveness of endogenous antioxidant system is diminished during aging, the exogenous supplementation of antioxidants might be a protective strategy against age-associated skin oxidative damage. It can be concluded that oxidative stress is a problem of skin cells and endogenous as well as exogenous antioxidants could play an important role in decreasing it. However, it is important to pretreat the skin with antioxidants before sun exposure. Animal and human studies have convincingly demonstrated pronounced photoprotective effects of “natural” and synthetic antioxidants when applied topically before UVR exposure. No significant protective effects of melatonin or the vitamins when applied alone or in combination were obtained when antioxidants were applied after UVR exposure. UVR-induced skin damage is a rapid event, and antioxidants possibly prevent such damage only when present in relevant concentration at the site of action at the beginning and during oxidative stress [42]. Treatment of the skin with antioxidants after the damage was caused by UVR might cause additional harmful effects on cell cycle control and apoptosis process. Antioxidants may thus have dichotomous activities with respect to carcinogenesis, namely, suppressing carcinogenesis by preventing oxidative damage to DNA [43] and promoting carcinogenesis by allowing survival of cells that are metabolically impaired (e.g., in altered matrix environments). Besides, the photoprotective effects of antioxidants are significant when applied in distinct mixtures in appropriate vehicles. According to Stahl et al. [23], endogenous photoprotection is complementary to topical photoprotection, and these two forms of prevention clearly should be considered mutually exclusive. The most important strategy to reduce the risk of sun UV radiation damage is to avoid the sun exposure and the use of sunscreens. The next step is the use of exogenous
5 antioxidants orally or by topical application and interventions in preventing oxidative stress and in enhanced DNA repair.
