Application note Demonstrating robust scale-up, packing consistency, and real-world bioprocess performance using the Evolve®D pre-packed column platform This application note demonstrates the scalable packing and operational performance of MiMode PuraBead® CX2 within the Evolve®D pre-packed chromatography platform across a range of process-scale formats. The study highlights consistent packed-bed efficiency, reliable hydrodynamic performance, and low operational backpressure during scale-up, while additionally demonstrating compatibility with commercially available third-party agarose-based affinity resins. Real-world process applicability was further evaluated using EtoxiClear® endotoxin removal workflows across development and manufacturing-scale columns. The data demonstrates the capability of the Evolve®D platform to support flexible, scalable, and manufacturing-ready chromatography processes across diverse downstream purification applications.

Introduction As downstream bioprocessing operations continue to increase in complexity and throughput, chromatography hardware and packing methodologies must deliver reproducibility, scalability, and dependable operational performance across a wide range of resin chemistries and process conditions. Single-use, pre-packed chromatography columns are increasingly being adopted to reduce process downtime, eliminate cleaning validation requirements, and improve manufacturing flexibility. However, maintaining consistent packed-bed performance across multiple resin platforms remains a significant challenge, particularly when scaling processes. The Evolve®D chromatography platform from Astrea Bioseparations has been developed to address these challenges through a scalable, process-ready, pre-packed column solution capable of supporting a broad range of chromatography media, including highly cross-linked agarose, mixed-mode, affinity, and specialty purification resins. In addition to its standard platform offering, we also provide custom column packing services using third-party resins, supporting customers from early-stage process development through to manufacturing-scale implementation. This application note demonstrates the scalable packing and operational performance of MiMode PuraBead® CX2 within the Evolve®D platform, alongside representative examples of additional agarose-based chromatography matrices. The study highlights the capability of the Evolve®D platform to maintain consistent hydrodynamic performance, reproducible packing efficiency, and low operational backpressure across multiple process-scale formats.

In addition to packing studies, this work includes real-world process performance data using Astrea Bioseparations’ EtoxiClear® endotoxin removal technology to demonstrate practical implementation under customer-relevant manufacturing conditions. Collectively, the data demonstrates the ability of the Evolve®D platform to support scalable, manufacturing-ready chromatography workflows across diverse downstream purification applications.

Evolve®D chromatography platform overview Evolve®D columns are advanced single-use chromatography systems developed to support the evolving demands of modern biopharmaceutical purification processes. Available process formats include column diameters ranging from 7 cm up to 60 cm, with a broad and flexible range of bed heights to support both development-scale and manufacturing-scale applications (Figure 1). The platform is designed to enable seamless scalability while maintaining consistent packed-bed hydrodynamic performance and robust operational reliability. The platform combines scalable packed-bed performance with a fully non-metallic flow path architecture (Figure 2), reducing the risks associated with corrosion, contamination, and biomolecule inactivation. By eliminating the need for column cleaning and validation, Evolve®D columns streamline manufacturing operations, reduce process downtime, and improve overall operational efficiency.
